What's the main difference between "leave", "day-off" and "vacation?" Or are they all interchangeable?

Not necessarily. These terms are not always interchangeable. In the work context, "leave" and "time/day-off" mean basically the same thing. If you want to say that you have time off work, then you can say that you have "leave" or a "day off." Ex: I have a day off tomorrow. Ex: I have two days leave from Thursday to Friday. Vacation can be used to describe extended leave or outside of the work context, just to describe personal leisure time in general. Ex: I will be taking two weeks vacation in June. Ex: I can't wait for summer vacation this year.
