I don't understand "I could sure use" part. How can I understand this expression?

"I could sure use some help ... " can be understood as "I could definitely use some help ... " whereas "sure" could be understood as "definitely." The grammar pattern "I could sure use [something]" is informally used to mean that you need or would like something. Ex: This website is too simple and boring. It could sure use an upgrade. Ex: Wow, it's so hot today! I could sure use a swim in the pool right now.
