"Make the most of it" is a common expression that means to enjoy something as much as you can. Whether that's high school, a job, a relationship, a bad situation, etc. you are actively trying to enjoy it. When he says "it" he means the students' high school experience. He wants them to enjoy it regardless of whether they have had good experiences or bad experiences in the past. Ex: I want you to make the most out of college. It's difficult but it is a lot of fun. Ex: You need to make the most out of life. Life is too short to not enjoy it.
