What does "not sure" mean? Is it okay to not put "I'm" at the front? And when can I use this expression?

"Not sure" is just a very casual, short form of "I'm not sure". "I'm not sure" means we do not know something for certain. Ex: I'm not sure what time we will get there. Ex: I'm not sure what this is.
