Good question! "A ton of" and "tons of" have the same meaning and can pretty much be used interchangeably. The main difference is that "a ton of" is normally used for countable nouns, and "tons of" for uncountable nouns. However, native speakers often mix these up all the time so this isn't always the rule. "Tons of" is also a little more casual and informal than "a ton of". Ex: Are you hungry? I have tons of food in the refrigerator! Ex: She's read a ton of books.
