It seems that the video has been cut or edited in a way that we have lost the "the" before "most important thing". "Most important thing" and "the most important thing" are the same things. In this particular sentence, grammatically, she has to have said the most important thing in order to be grammatically correct. Ex: The most important thing is to always smile. Ex: Never forget the most important thing, which is to be happy.
