What does "reply" mean in the debate context? I thought it is only used to messaging context.

Great question. "Reply" in a debate context means a rebuttal, to counteract and to argue the other side of the debate; in this case being against immigration policies. The teacher is asking Amber to reply and counteract what was said by Cher in the debate. Of course, reply is more often used in a messaging context than it is in a debate context, but it still can be used in that way.
