"In one's pursuit" in this video means to work for something; to do an activity regularly to achieve a larger goal. It can also mean to chase. Tim Grover, Kobe Bryant's trainer, is saying that Bryant worked out constantly and practiced frequently to achieve his goal of becoming an excellent basketball player.
