"In one's pursuit" in this video means to work for something; to do an activity regularly to achieve a larger goal. It can also mean to chase. Tim Grover, Kobe Bryant's trainer, is saying that Bryant worked out constantly and practiced frequently to achieve his goal of becoming an excellent basketball player. Ex: In the Declaration of Independence, it says we have the right to life, liberty, and the pursuit of happiness. Ex: Meditation helps me in my pursuit of mindfulness.
