What does "lady of the house" mean? Does it simply refer to the house owner? If so, could I use it for a male as well?

The "the lady of the house" refers to the most important or responsible female in the house. Such as a wife, mother, or if only a lady lives in the house. You can use this phrase with males as well! You can say "the man of the house". The phrase isn't common to say these days, but you can use it! Ex: After dad left, I became the man of the house. Ex: Where is the lady of the house? Is she coming to the party?
