Can I say "how may I help you?" as well instead of "how can I help you?" Is there a difference?

Yes, of course you can! They mean the same thing, but using "may" is slightly more formal in tone than "can." Ex: May I help you? (Formal) Ex: Can I go to the washroom? (More casual)
