"I'm up" here is in reference to mood. Being up means to be in a happy/good or positive mood. It is the contrast to the phrase "feeling down" which means to feel sad or some other negative emotions. It is not commonly just used by itself. So I'm up would not be a natural reply to someone asking how you're feeling. It is most often used, like in this clip, as a contrast to down. Ex: My mood had been really up and down lately. Ex: What's the matter? You seem very up and down today.
