There isn't a huge difference between the two, but you are correct in thinking that "puke" is more informal and casual than "vomit," which is used in more formal situations. Another expression similar to "puke" is "throw up." Ex: The new patient is vomiting and requires medical attention. Ex: I felt sick after eating the burger, so I puked it all out.
