Does this sentence have two different sentences combined? Like "what do you think?" and "what does make a good leader?" Is this a correct sentence?

Not quite! The sentence is correct, but it just has a couple of extra words and phrases in it. In its simplest form, it'd probably be "What makes a good leader?" the addition of "do you think" lets us know that he's asking about their opinions and thoughts on the subject. It's similar to asking, "what's your opinion on...?" Ex: What did you think about the baseball game? Ex: What do you think makes someone a good person? = In your opinion, what makes someone a good person?
