The difference is "emphasis!" "single" adds more emphasis to the phrase. "Every week" is more neutral. So if you want a slightly dramatic tone or to emphasize that it happens "every week," you can add "single" to it. Ex: I sit behind a desk every single week and look forward to the weekend. Ex: I do swimming practice every week. Ex: He says that every single week! It's annoying.
