"Hold on" in this context means to wait. Harry says hold on because he is telling Ron to wait to speak. This is because he hears the train whistle and wants to listen to it. "Hold on" can also mean to grab and cling to something tightly. Ron says it to Harry later on in this video because he is falling out of the car and is gripping to the car door.
