No, there really isn't a difference between these two phrases. "How much time have I got?" grammatically, is incorrect. "How much time do I have?" is the grammatically correct way to say this question. However, both are common to use, so yes, you definitely can use this sentence instead of the one that is used in the video. Ex: A: How much time do I have? B: You have 15 minutes. Ex: A: How much time have I got? B: You have 15 minutes.
