"Burden" is much heavier in tone and has a more negative connotation than "responsibility" does. A "burden" is considered to be something unpleasant, distressing, and a generally unwanted responsibility. "Responsibility" doesn't necessarily carry the same negative connotations. Ex: My assistant has the responsibility of filing all my work documents. Ex: The young man had the burden of caring for his ill parents and all of his siblings.
